Sorghum bicolor supplement contains various phytochemicals, including phenolic acids and polyphenols such as proanthocyanidins. Sorghum bicolor supplement is particularly rich in 3-deoxyanthocyanins, such as luteolinidin and apigeninidin, and appears to induce apoptosis and inhibit cell proliferation in cancer cells through the stimulation of various apoptosis promoter genes and the downregulation of certain apoptosis inhibitor genes. In addition, due to the strong antioxidant nature of the phytochemicals, these compounds are able to scavenge free radicals and prevent tissue damage. Also, intake of this supplement modulates the immune system by both increasing the activity of natural killer (NK) cells and initiating the activation of macrophages.<\/div>\n<figure Cell viability, tumor necrosis factor- (TNF-) \ud835\udefc, and interleukin- (IL-) 6 were measured in RAW 264.7 macrophages treated with increasing doses (0,15, 30, and 60 \ud835\udf07g\/mL) of sorghum ethanol extracts and stimulated with lipopolysaccharide (LPS). Extract dose had a significan effect on TNF-\ud835\udefc and IL-6, with a trend of cytokines decreasing between 0 \ud835\udf07g\/mL and 15 \ud835\udf07g\/mL of sorghum extract. Genotypealso had a significant effect on the cytokines, with extracts from four accessions significantly decreasing TNF-\ud835\udefc and\/or IL-6.<br \/>\nCells treated with 3-deoxyanthocyanidin-containing accessions had less cytokine production than non-3-deoxyanthocyanidin accessions, whereas cells treated with proanthocyanidin-containing accessions had more cytokine production than cells treated with nonproanthocyanidin accessions. Additionally, there was a significant effect of the Tannin1 allele on TNF-\ud835\udefc and IL-6. Our results demonstrate that sorghum genotypes differentially modulate induction of inflammatory cytokine production in RAW 264.7 macrophages and that specialty grain has the potential to be tailored by controlling traits at the nucleotide level.
